La leucémie lymphoďde chronique des lymphocytes B (LLC-B) est caractérisée par une remarquable déficience en apoptose. Nous avons cherché ŕ savoir si l'hyperforine, dérivé de la plante Hypericum perforatum, pourrait induire l'apoptose des cellules leucémiques de patients atteints de LLC-B ex vivo. Les résultats montrent que le traitement des cellules de LLC-B déclenche un certain nombre d'événements apoptotiques : externalisation de la phosphatidylsérine, fragmentation des noyaux, activation de la caspase-3 et clivage d'un substrat de caspase, la PARP, ainsi que la régulation négative de Bcl-2 et Mcl-1, deux protéines anti-apoptotiques. De plus, l'hyperforine régule négativement deux autres protéines surexprimées par les cellules de LLC-B: la NO synthase de type II (iNOS) qui catalyse la production du NO et qui est également anti-apoptotique dans ce modčle, ainsi que la p27kip1, un inhibiteur des kinases dépendantes des cyclines. En conclusion, l'hyperforine semble ętre un composé naturel prometteur pour rechercher de nouvelles stratégies thérapeutiques des LLC-B basées sur l'induction d'apoptose et sur la combinaison avec des agents antitumoraux dépendants du cycle cellulaire.
